Qualifier Jenson Brooksby withdrew after the main draw was published due to abdominal pain, and was replaced by lucky loser Dominik Koepfer.

- Marton Fucsovics defeated Fabio Fognini 6-1 6-7(6) 7-6(5)
- Adrian Mannarino defeated Nikoloz Basilashvili 6-2 6-4
- Cameron Norrie (10) defeated Federico Delbonis 6-2 6-1
Norrie remains one of five players in contention for the two places left in the ATP Finals draw
- Lucky loser Lorenzo Musetti defeated Laslo Djere 4-6 7-6(5) 6-4
- Lucky loser Dominik Koepfer defeated wildcard Andy Murray 6-4 5-7 7-6(9)
Murray failed to convert 7 match points and Koepfer won the contest after converting his first.
- Alexander Bublik defeated Daniel Evans 2-6 7-5 7-5
- Dusan Lajovic defeated Mackenzie McDonald 6-3 6-4
- Qualifier Hugo Gaston defeated wildcard Arthur Rinderknech 4-6 6-4 6-3
This win will see Gaston move into the top 100 for the first time
- Pablo Carreno Busta (12) defeated Benoit Paire 6-3 6-4
- Sebastian Korda defeated Aslan Karatsev (13) 6-2 6-7(9) 7-6(5)
After winning the title in Moscow, Karatsev has lost his opening match in the next two tournaments, St. Petersburg and here in Paris.
